Karmanos scientists join Welsh, Egyptian colleagues to contribute cover story to a major AACR journal

 
 

Scientists from the Barbara Ann Karmanos Cancer Institute, teaming with colleagues from Wales and Egypt, have published an article on the identification of a small molecule inhibitor that could thwart development and progression of breast cancer. The article serves as the cover story in the April edition of “Molecular Cancer Therapeutics,” a major journal of the American Association for Cancer Research.

In the article, the team led by Dr. Malathy Shekhar, Ph.D., associate professor of Oncology at Wayne State University School of Medicine (WSU SOM) and the Molecular Therapeutics Program of Karmanos Cancer Institute (KCI), and Dr. Andrew Westwell, Ph.D., from the School of Pharmacy and Pharmaceutical Sciences at Cardiff University, UK, describe their work in the development of the first small molecular inhibitor for Rad6.

Rad6 is a cellular protein that plays a critical role in the development and spread of breast cancer. By targeting Rad6, the small molecule inhibits breast cancer cell survival and growth. Rad6 functions by enabling destruction of important cellular proteins via proteasomes, a major protein degradation system.  Drugs targeting the proteasome have been tested in clinical trials. However, their efficacy is limited due to side-effects.  By targeting Rad6, this novel inhibitor could provide greater specificity and be potentially more effective in treating cancers, according to the published research.
